Walk down any supplement aisle and the skin products tend to share three problems.

Here is the distinction that changes how you should read a skin label. Collagen is assembled from amino acids, and the marketing logic follows easily: supply more amino acids, build more collagen. But in someone eating adequate protein, collagen synthesis is rarely limited by amino acid supply. Lysine and proline are abundant in ordinary diets. What synthesis is commonly limited by are the cofactors — and those are far more often insufficient.

Vitamin C is the clearest example. A systematic review in the Orthopaedic Journal of Sports Medicine[7] describes vitamin C's role as the required cofactor for prolyl hydroxylase and lysyl hydroxylase — the enzymes that hydroxylate proline and lysine residues so the collagen triple helix can fold into a stable structure. Without it, the collagen your body assembles is structurally unsound. That review examined musculoskeletal healing rather than skin, but the enzyme chemistry is identical in both tissues. Biotin: A Smaller Claim Than the Category Makes

Biotin 2,500 mcg is a B-vitamin involved in keratin production, the structural protein in skin, hair, and nails. Deficiency produces brittle nails and hair thinning. Supplementation in someone already replete does considerably less than the category implies, and we would rather say that than imply otherwise. It is a reasonable inclusion at a sensible amount, not a miracle.

One practical warning that genuinely matters. Biotin can interfere with laboratory tests that use biotin-streptavidin technology, which includes many thyroid panels and cardiac troponin assays, producing falsely high or falsely low results depending on the test. The NIH Office of Dietary Supplements notes[13] that supplementing beyond recommended intakes can cause clinically significant incorrect results, and cites research finding that a single 10 mg oral dose interferes with thyroid function tests. The FDA maintains a list of troponin assays subject to this interference[14], having flagged a case where a falsely low troponin result contributed to a patient death.
